DH5a Escherichia coli Strains Description

DH5 Alpha strain is the most commonly used strain in the laboratory. The deletion of nucleic acid endonuclease (endA) increased the yield and quality of plasmid DNA, and the recombinant enzyme deficiency type (recA) reduced the homologous recombination probability of the inserted fragment and ensured the stability of the insertion of DNA; DH5 alpha was combined with the LacZa polypeptide produced by the carrier DNA and lacZ Delta M15 phase encoded by DH5 a when the pUC series plasmid vector was used for DNA transformation. Thus, the activity of beta galactosidase (alpha complementarity) was displayed, so that DH5 could be used for screening blue and white spots. DH5 can be used to make gene library and subclone, and because DH5 alpha has deoR variation, it can be used as the host plasmid of larger plasmid. The disadvantage of the DH5 strain is slow growth, and it takes 12-14 hours at 37 C to see cloning.
