GS115 Pichia pastoris Yeast Strain Description

GS115 is a Pichia pastoral strain, which belongs to eukaryotic cells. According to the general antibiotics such as prokaryotes and yeast to ampicillin kanamycin is invalid, so in order to prevent Escherichia coli and other prokaryotes strains pollution on yeast, often in the cultivation of adding some ampicillin and kanamycin antibiotic medium to inhibit bacterial growth and contamination. The phenotype of Pichia Pichia GS115 is Mut+, but the GS115 transformant can produce both Mut+ strain and Muts strain. The expression level of target protein in these two transformation lines may be different and unpredictable, so only by experiment can we get the best yeast expression plan.

The suitable growth temperature of Pichia pastoris is 28 to 30 degrees, and the temperature exceeding 32 degrees is harmful to the expression of protein and may lead to cell death. GS115 Pichia pastoris is histidine deficient type (His4 genotype). If histidine gene is carried on the expression vector, it can compensate histidine deficiency of host bacteria, so it can be screened on transformants without histidine. The probability of the spontaneous mutation of these receptor bacteria to the histidine wild type is generally less than 10-8. Pichia pastoris GS115 can grow in YPD medium or grow in minimal media supplemented with histidine, but it can not grow in a single minimal media medium.

Pichia pastoris, a type of yeast that can use methanol as the only carbon source and energy, is a kind of methanol nutritive yeast. Like other yeasts, it exists mainly in haploid form at asexual growth stage. When environmental nutrition is limited, 2 physiological types of haploid cells are induced to mate and fuse into diploid. Another biological characteristic of Pichia Pichia Pasteur is that the alcohol oxidase needed for methanol metabolism is sorted into peroxisomes to form regionalization. When glucose was used as carbon source, there were only 1 or few small peroxisomes in the cell. When methanol was used as carbon source, peroxisome accounted for almost 80% of the total cell volume, and AOX increased to 35%-40% of total protein. Therefore, when the AOX gene is inserted into the foreign protein gene by homologous recombination, a large amount of expression can be obtained. At the same time, according to the formation of methanol yeast peroxisomal characteristics, not only can use the system to express some toxic protein and susceptible to degradation enzymes, may also occur and its mechanism and function of cell specific regional organisms provide inspiration for similar research in higher animal.
